Colon Cancer Tissue Microarray (TMA)

  • Designed for examination of associations of markers with tumor stage, clinical outcome, and other clinico-pathologic variables in Stage I – IV (Data Manual).

  • The TMA specimen set consists of
    • Invasive colon carcinoma
      - Four stages: N = 367
    • Normal colon epithelium
      - Adjacent to the cancer: N = 34
      - From diverticulitis patients with no cancer: N = 40
    • Adenomatous polyps, no cancer: N= 37
    • Controls
      - Normal non-colon tissues: N = 32
      - Cell lines: N = 48

  • Four replicate TMA blocks are created from each case set
    • Four cores are taken from each case block, with one of those cores placed in each of four replicate array blocks.
    • Example of a Colon TMA map. Actual maps are supplied with the TMA shipment.

  • The TMA was designed by NCI statisticians for high statistical power for comparisons of marker values:
    • Between colon cancers of different stages
    • Between colon cancers that recurred within a few years of diagnosis versus colon cancers that remained recurrence free.

    For example, there is 80% power to detect:
    • Difference in prevalence rate for a binary marker of about 0.30 between stage I and stage IV cancer
    • Difference in prevalence rate of about 0.35 between stage II colon cancers that recurred within 5 years versus those that did not recur within 5 years.


  • Information for invasive colon carcinoma cases:
    • Race, gender and age at diagnosis
    • Year of diagnosis
    • Primary site within colon
    • TNM stage, number of positive nodes and total examined
    • Initial treatment—only Y/N: chemotherapy, radiation, surgery
    • Histology, grade, lymphovascular invasion, status of margins
    • Recurrence status and time to recurrence
    • Vital status and time to death
    • Cancer status at last follow-up

  • Information for adenomatous polyp cases:
    • Gender
    • Age at diagnosis
    • Year of diagnosis
    • Polyp type
    • Degree of dysplasia
    • Gross size of polyp
